What you would need to provide
pAAV-ITR-GOI-ITR DNA prep:
- Size ≈ 4.7kb
- Endonuclease & recombination deficient competent cells (SURE, Stbl3)
- Endotoxin-free maxi prep
- Control cut with SmaI restriction enzyme
- 200ug DNA for small scale prep, 500ug for large scale prep
AAV capsid DNA prep:
- Endonuclease & recombination deficient competent cells (SURE, Stbl3)
- Endotoxin-free maxi prep

Consultation on vector design
Two production scales
- Small scale (½ prep): 500ul
- Large scale (full prep): 1ml
Expected yield*
- AAV: 1ml of ≥ 1x1013 vg/ml
- Lentivirus: 1ml of ≈108 TU/ml
*Some transgenes can be cytotoxic, and occasionally different capsids can produce less efficiently. This can result in lower titer.

Lentivirus

Users can decide to pay 10% extra charge as insurance for failed preps. This will allow users to run an extra production process without additional costs.
