We found that reduced activity of RNA polymerase I extends healthy lifespan in Drosophila. This lifespan extension can be recapitulated by silencing Pol I exclusively in the cell-types of the adult midgut. Reducing Pol I activity improves multiple aspects of organismal health, delaying age-related decline in neuromuscular function and preventing age-related gut hyperplasia.
