|Module aims:||Introduction to techniques and approaches that are used to investigate biochemical problems, together with their underlying principles. It is also designed to give experience in practical work, calculations and reading scientific papers.. Specific areas are pH, redox potentials and equilibrium; basic spectroscopy; amino acids, protein sequences and structures; mass spec., NMR and X-ray methods; statistical methods; protein purification and assays; molecular biology methods; metabolic pathways; uses of isotopes in biochemistry.|
|Module objectives:||• be able to make straightforward biochemical calculations, at a level of those included in the tutorial exercises • understand pH, buffers, chemical equilibrium, exergonic and endergonic reactions, acid dissociation constant (pKa) and redox potentials • understand the basics of UV/visible and fluorescence spectroscopy and their applications in biochemistry • become familiar with the amino acid components of proteins, how they link together and how you can determine amino acid sequences of proteins • understand how amino acids form different types of protein structures and the different methods available for the determination of such structures • appreciate the essentials of experimental design and data presentation, statistical assessment and interpretation • know the main methods of protein purification and characterisation, and the properties of proteins on which these are based • appreciate the uses of molecular biological techniques in biochemical research • understand the use of polyclonal and monoclonal antibodies for detection and quantification of biological molecules • gain an overview of major metabolic pathways in intact cells and tissues • understand the use of isotopes in Biochemistry • be able to read and understand a scientific paper • gain practical experience in spectrophotometry, enzyme assay and column chromatography|
