Our general approach is to apply structural techniques including protein crystallography and crosslinking coupled to mass spectrometry to gain insights into the structure of protein complexes that respond to second messengers during synaptic plasticity. This enables us to generate novel functional hypotheses that we test using methods including patch clamp electrophysiology with rat brain slices. In this way, we tie structure to function. We are particularly interested in understanding how anchoring proteins target cAMP-dependent protein kinase and calcineurin during long-term depression, and calmodulin-dependent protein kinase II (CaMKII) during long-term potentiation.
