Research Summary

COLORECTAL CANCER: We have delineated cellular and molecular mechanisms and mapped overexpression of a growth promoting peptide (Endothelin-1) and its receptor (Endothelin A receptor). We evaluated endothelin A receptor antagonism as a novel treatment agent and demostrated cancer killing. Agents (against endothelin A receptors) are being entered in clinical trials of patients with advanced colorectal cancer. The aim is to provide effective drug treatments for advanced colorectal cancer. MULTIDRUG RESISTANT CANCERS (breast and bladder): We have described a novel drug delivery system: photochemical internalisation of chemotherapy. This involves delivery of chemotherapeutics in an endosome whose membrane contains photodynamic agents. The latter are activated upon illumination resulting in the endosomal membrane bursting and chemotherapeutic agent delivered intracellularly. Other nanoscale drug delivery systems are also under investigation. The aim is to bypass drug-resistance which tends to develop in the majority of solid cancers. REGENERATIVE MEDICINE: We are constructing a nanopolymer based scaffold for the small intestine. The scaffold incorporates different cell types and will encourage nerve regeneration and the production of a blood supply. The ultimate aim is to tissue engineer tissue to treat small bowel syndrome.
