Research Description

The focus of my research interest is the role of macrophages as sentinel cells of innate immunity which activate and regulate host immune responses to wide ranging microbial stimuli, and also function as host defence effector cells by intracellular killing of pathogens. In particular, I study host cell interacions with HIV-1, which can infect and actively replicate in marophages without evident cytopathic effect. In a model of HIV-1 infection of primary human macrophages I test host cell innate immune responses to HIV-1 itself, the effect of established HIV-1 infection on responses to clinically important co-infecting pathogens and to interferons that are important regulators of macrophage effector function. These studies have led to collaboration with Greg Towers, on mechanisms of HIV-1 restriction in macrophages, and with Richard Jenner on the effects of HIV-1 Tat in macrophages. In addition I am engaged in collaborative studies to investigate macrophage responses to Mycobacteria and in particular to develop research methodology to measure differential human macrophage and T cell responses to Mycobacterial stimulation in vivo.

I have developed a specific interest in measurement of the signalling pathways that underpin innate immune responses. This work has led to successful adaptation of a quantitative confocal microscopy assay for nuclear translocation of key transcription factors (NFkB, IRF3 & IRF7)at single cell level, with widespread applications in Infection & Immunity research, leading to an ongoing collaboration with Mary Collins on NFkB activation by the viral FLIP protein of KSHV. A further extension of this interest, has led to collaborative studies with Benny Chain, to develop of a clinical assay for quantitative measurement of innate immune cellular activation in order to screen for inherited or acquired abnormalities of cellular innate immune signalling pathways that are becoming increasingly evident.
