Matrix-Assisted Laser Desorption Ionisation Quadrupole-Time of Flight Mass Spectrometry (MALDI QTOF MS) and Electrospray Quadrupole - Time of Flight Mass Spectrometry (ESI-QTOF)
The Unit benefited from a SRIF award that involved the procurement of a dual-source (MALDI and ESI) time-of-flight mass spectrometer. MALDI QTOF MS is a very sensitive ionisation technique that is used primarily within the unit for identification of proteins excised from gels and the analysis of carbohydrate structures.
ESI QTOF MS is a dynamic
technique that allows the coupling of the mass spectrometer to a
liquid
chromatography interface (nanoLC). ESI QTOF MS is used for
the sequencing / identification of proteins and / or locating
post-translational
modifications such as phosphorylation / glycosylation /
alkylation
etc. The
ESI QTOF MS is also used in conjunction with 2-dimensional
liquid chromatography
to study changes in protein expression as a complement to
existing
2D-PAGE analyses.

Typhoon Trio Scanner (2D-PAGE / 2D-DIGE)
The department also has a Typhoon fluorescent scanner which is used for 2D-DIGE analysis and for detecting quantative changes in protein expression.
2D-PAGE / 2D-DIGE
The Centre has several IPG-PHOR
isoelectric focusing, 1D and 2D-PAGE electrophoresis units
including
a Protean II Mulitcell and Ettan Dalt-12 gel systems. There is
also a Safe Imager (blue light transilluminator) for viewing
ultra-violet
sensitive stains (Sypro Ruby, Deep Purple etc.).

Nano-High Performance Liquid Chromatography Unit
Solvent and sample delivery to the ESI-Q-TOF is performed using a NanoAcquity HPLC

The Centre has recently acquired tissue imaging capability
